The Spike (SARS-CoV-2, UK variant) Pseudotyped Lentivirus were produced with SARS-CoV-2 UK Variant Spike (Genbank Accession #QHD43416.1 with UK variant mutations; see below for details) as the envelope glycoproteins instead of the commonly used VSV-G. These pseudovirions contain the firefly luciferase gene driven by a CMV promoter, therefore, the spike-mediated cell entry can be measured via luciferase activity. The Spike (SARS-CoV-2, UK variant) pseudotyped lentivirus can be used to measure the activity of neutralizing antibody against SARS-CoV-2 UK variant in a Biosafety Level 2 facility.

PIM (ABS) – Human AB Serum is a required additive for PIM(R), PIM(S), and PIM(T) islet specific tissue culture media. It is shipped separately as a frozen component to permit the extended shelf life of this media without the AB Serum. Once it is thawed and added to the media, it is recommended that the expiration date be considered to be 60 days from the date of the PIM(ABS) addition. It is collected, screened, and frozen until ready for cGMP manufacturing which includes heat inactivation followed by sterile filtration and bottle filling. All lots of PIM(ABS) are screened for compatibility with human islets prior to release in addition to the standard QC/QA testing and analysis.

PIM(S)® is a new Prodo Labs islet specific media that is manufactured under cGMP conditions for use in human islet culture for clinical transplantation and for research. It is designed for long term tissue culture of islets from 2 to 21 days at 37oC. It is enhanced with specific components that permit long term culture of healthy islets that maintain their glucose responsiveness, insulin content, and suitability for implantation. It currently is designed to be used with 5% Human AB Serum that is pretested for islet suitability. It does not contain any animal proteins or growth factors and has a glucose concentration of 5.8 mM.

The one component 3,3?,5,5?-tetramethylbenzidine (TMB) substrate is supplied in a ready-to-use solution. Peroxidase reacts with the TMB substrate solution to produce a soluble blue reaction product that can be read at 370 nm or 620-650 nm for ELISA applications.
