The Notch CSL Reporter - HEK293 cell line contains the firefly luciferase gene under the control of Notch-response elements (CSL responsive elements) stably integrated into HEK293 cells. Transfection of this cell line with a Notch expression vector and activation of the Notch pathway leads to expression of luciferase reporter. This cell line is validated with a Notch1DelE expression vector, which is constitutively processed by γ-secretase, leading to the release of NICD and the expression of luciferase.

The BCMA / NF-κB - HEK293 recombinant cell line has been transfected with full-length human BCMA cDNA (Genbank #NM_001192) under control of a CMV promoter for high constitutive expression. The NF-κB-luciferase reporter is also stably integrated into the genome. The firefly luciferase gene is controlled by four copies of the NF-κB Response Element upstream of a minimal promoter. Upon ligand binding, active BCMA will initiate the NF-κB signaling pathway, leading to expression of the NF-κB-controlled luciferase reporter.

It contains a firefly luciferase gene driven by the STAT5 response element located upstream of the minimal TATA promoter. After activation by cytokines or growth factors, endogenous STAT5 binds to the DNA response elements, inducing transcription of the luciferase reporter gene.

It is made from the anti-CD19 CAR lentivirus (BPS Bioscience #79851). The reporter cell line has been validated for anti CD19-CAR expression by FACS, and for luciferase reporter activation stimulated by target cells including CD19/CHO recombinant cell line can be used for primary screening and functional validation of anti-CD19 CAR construct and lentivirus before testing in primary T cells.<br /><br />Anti-CD19 CAR consists of anti-CD19 scFv linked to 3rd generation CAR (Chimeric Antigen Receptor) containing CD28, 4-1BB co-stimulatory domains, and CD3ζ signaling domain.<br />

Anti-CD19 CAR negative control/NFAT-luciferase reporter Jurkat cell line is a double stable cell line expressing anti-CD19 CAR negative control and NFAT-luciferase reporter. The anti-CD19 CAR negative control consists of anti-CD19 scFv linked to the CD28 transmembrane motif without any intracellular signaling domains. The reporter cell line has been validated for anti- CD19 expression by FACS, while the stimulation by target cells including CD19/CHO recombinant cell line has not activated the luciferase reporter gene in this cell line. The cell line can be used for the negative control of anti-CD19 CAR/Jurkat-NFAT cell line (BPS Bioscience, #79853).
